چکیده

Burkholderia glumae produced antibiotic substance(s) on agar plate media, but none in various liquid media. Some minor elements in agar were considered to be involved in the antibiotic production. K+, Ca'~, Mg" and NH4 ~ were shown to play significant roles in the production by culturing the bacterium in liquid media supplemented with various inorganic chemicals. Highest production was observed when B. glumae was cultured in the liquid medium containing a mixture of MgCI" KCl and CaCI,. No antibiotic production was observed in PGC broth containing MgS04which was substituted for MgCI, or Mg(NO"h These results suggest that specific cations and anions are involved in antibiotic(s) production by B. glumae.

Dissection of Quorum-Sensing Genes in Burkholderia glumae Reveals Non-Canonical Regulation and the New Regulatory Gene tofM for Toxoflavin Production

Burkholderia glumae causes bacterial panicle blight of rice and produces major virulence factors, including toxoflavin, under the control of the quorum-sensing (QS) system mediated by the luxI homolog, tofI, and the luxR homolog, tofR. In this study, a series of markerless deletion mutants of B. glumae for tofI and tofR were generated using the suicide vector system, pKKSacB, for comprehensive ...

Diversities in Virulence, Antifungal Activity, Pigmentation and DNA Fingerprint among Strains of Burkholderia glumae

Burkholderia glumae is the primary causal agent of bacterial panicle blight of rice. In this study, 11 naturally avirulent and nine virulent strains of B. glumae native to the southern United States were characterized in terms of virulence in rice and onion, toxofalvin production, antifungal activity, pigmentation and genomic structure. Virulence of B. glumae strains on rice panicles was highly...

Quorum Sensing Controls Swarming Motility of Burkholderia glumae through Regulation of Rhamnolipids

Burkholderia glumae is a plant pathogenic bacterium that uses an acyl-homoserine lactone-mediated quorum sensing system to regulate protein secretion, oxalate production and major virulence determinants such as toxoflavin and flagella. B. glumae also releases surface-active rhamnolipids. In Pseudomonas aeruginosa and Burkholderia thailandensis, rhamnolipids, along with flagella, are required fo...

BIOTECHNOLOGICALLY RELEVANT ENZYMES AND PROTEINS Mutations improving production and secretion of extracellular lipase by Burkholderia glumae PG1

Burkholderia glumae is a Gram-negative phytopathogenic bacterium known as the causative agent of rice panicle blight. Strain B. glumae PG1 is used for the production of a biotechnologically relevant lipase, which is secreted into the culture supernatant via a type II secretion pathway. We have comparatively analyzed the genome sequences of B. glumae PG1 wild type and a lipase overproducing stra...

Effect of single additive bivalent metal ions on the growth of Streptomyces clavuligerus and clavulanic acid production

Metal ions are the main impurities of water and media ingredients used in fermentation processes. In this research, the effect of Ca+2, Co+2, Cu+2, Fe+2, Mg+2, Mn+2 and Zn+2 as chloride and sulfate salts was studied on the clavulanic acid production and Streptomyces clavuligerus growth. All chloride salts had negative effect on clavulanic acid production and no clavulanic acid was produced in t...
